The Anti-Aging Supplement Loved by Celebrities

In recent years, a staggering 50% decrease in nicotinamide adenine dinucleotide (NAD+) levels has been observed between individuals in their twenties compared to those in their sixties, spotlighting a critical aging marker [1].

The decrease highlights the effects of getting older. It explains why NAD+, an essential coenzyme in metabolic functions, has received a lot of interest, especially from Hollywood celebrities looking for ways to stay youthful.

What is NAD+?

NAD+ is essential for the fundamental processes that fuel our cells, aiding in everything from energy metabolism to DNA repair and cell signaling.

As we age, our bodies’ natural ability to produce NAD+ diminishes, leading to many age-related health issues.

NAD+ is synthesized from vitamin B3 and stimulates over 500 biochemical reactions, targeting enzymes crucial for cellular health [1], [2].

Dr. Charles Brenner, a biochemist and Chief Scientific Advisor of Chromadex, underscores its significance by stating, “NAD coenzymes are the central catalysts of all living things,” and explains further that, “They underlie the conversion of protein, fat, and carbohydrate into energy. They underlie the conversion of everything that we eat into everything that we do and everything that we are. They are required for our cells to generate energy” [4]

Boosting NAD+ levels has been linked to improved energy metabolism, DNA repair, and circadian rhythm regulation. This makes NAD+ a critical target for those looking to mitigate the effects of aging and enhance overall cellular function.

Research has linked increased NAD+ levels with improved muscle function, better glucose metabolism, and enhanced mitochondrial health [3].

These effects suggest that NAD+ could help manage general aging and conditions like diabetes and metabolic syndrome.

Studies have also indicated that NAD+ supplementation may help improve brain health, cognitive function, and recovery from neurodegenerative diseases such as Alzheimer’s [4].

Additionally, NAD+ has been shown to support liver health and aid in recovery from alcohol-related damage, showcasing its wide-ranging potential benefits.

The buzz around NAD+

Celebrities like Hailey Bieber and Jennifer Aniston have openly endorsed the supplement, claiming it boosts vitality and aids recovery.

“I’m going to NAD for the rest of my life, and I’m never going to age,” Hailey Bieber shared in a documentary, encapsulating the allure of NAD+ among those in the limelight [4].

Jennifer Aniston, known for her rigorous wellness routine, has praised NAD+ for its rejuvenating effects.

Podcaster Joe Rogan regularly features NAD+ discussions, emphasizing its role in maintaining peak physical and mental performance [1].

Scientific research

Despite the celebrity endorsements, what does the science say? According to research by Leonard Guarente, Ph.D., a professor at MIT, NAD+ plays a “central role in the generation of cellular energy” [2].

Studies have shown that NAD+ supplementation can increase the abundance of this coenzyme in the blood, correlating with improvements in aging-related biomarkers.

Clinical trials have demonstrated that NAD+ precursors like nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN) are safe and can effectively raise NAD+ levels in humans, showing promise in improving muscle function, metabolic health and reducing inflammation [1], [3].

Dr. David Sinclair of Harvard Medical School points out that while the initial data is promising, NAD+ supplementation is “not a magic pill” and should be part of a broader approach to health and wellness [1].

“It’s about optimizing body functions as part of an integrated approach to aging,” Sinclair notes.

He emphasizes the importance of combining NAD+ supplementation with lifestyle changes such as regular exercise, a balanced diet, and adequate sleep to maximize the potential benefits.

This holistic approach is supported by evidence showing that lifestyle factors can significantly influence NAD+ levels and overall health outcomes.

Cautions and considerations

However, the long-term effects of NAD+ supplementation are not fully understood, and the efficacy can vary dramatically between individuals [2], [3].

Experts like Dr. Sinclair advise, “It’s crucial to approach these supplements with a well-rounded perspective, integrating them with lifestyle changes and medical advice” [1].

Moreover, some studies have raised concerns about the bioavailability and effectiveness of various NAD+ delivery methods, such as oral supplements versus IV infusions, highlighting the need for further research to determine the most effective protocols.

The trajectory of NAD+ research is aimed at more robust clinical trials to solidify its benefits and establish standardized dosages and administration protocols.

The goal is to validate the anecdotal claims and understand the broader implications of long-term NAD+ supplementation [2].

Researchers are also exploring the potential of combining NAD+ precursors with other compounds to enhance their efficacy and address the underlying causes of NAD+ decline more effectively.

This multi-targeted approach could lead to more comprehensive anti-aging strategies and improved health outcomes for a wider range of individuals.
